The Lambton Worm in Low Row is one of more than 40 venues put on the market by operator JD Wetherspoon.
The pub is located on the ground floor of the Sunderland Central Travelodge Hotel.
The chain has appointed estate agents Savills and CBRE to handle the disposal of 45 of its pubs.
The majority are located in London and the South, but others in the North East include The Union Rooms in Newcastle and The Leaping Salmon in Berwick.
